Combined topical and systemic administration with human adipose-derived mesenchymal stem cells (hADSC) and hADSC-derived exosomes markedly promoted cutaneous wound healing and regeneration

Abstract Background Cutaneous wound healing and regeneration have become a recognized health challenge in the world, which causes severe damage to mental physical of patients. Human adipose-derived mesenchymal stem cells (hADSC) play an essential role via their paracrine function. Exosomes secreted by hADSC may contribute this progress. In study, we investigated potential clinical application roles hADSC-derived exosomes (hADSC-Exo) cutaneous healing. Methods hADSC-Exo was isolated from human ultracentrifugation. Mice were subjected full-thickness skin biopsy experiment treated with either control vehicle or smearing administration (sm) subcutaneous (sc) intravenous (iv). The efficacy on evaluated measuring closure rates, histological analysis. Results Combined local hADSC/hADSC-Exo offered additional benefit promoting healing, accelerating re-epithelialization, reducing scar widths, enhancing angiogenesis collagen synthesis. Either topical systemic appeared more effective stimulating cell proliferation, inhibiting apoptosis inflammation, elasticity barrier integrity, increased genes expression PCNA, VEGF, III, Filaggrin, Loricrin, AQP3, decreased TNF-alpha. Conclusion Our findings suggest that combined can facilitate reduce formation. These data provide first evidence for feasibility as cell-free therapy treating wounds, value hADSC/hADSC-Exo.
